Summary
Argireline (acetyl hexapeptide) is a topical cosmetic peptide marketed as a needle-free, 'botox-like' alternative that works by inhibiting neurotransmitter release at the neuromuscular junction [2]. Peptide cosmeceuticals as a class have a relatively strong evidence base in photoaging reviews, with many studies reaching higher levels of evidence [1], but direct human data for Argireline specifically are mixed: a controlled split-face study using objective Visia imaging found differences between treated and untreated sides were not statistically significant, concluding its anti-wrinkle effect was not demonstrated while noting low toxicity [3]. Supportive efficacy evidence is largely preclinical, such as improved skin histology and increased type I collagen in aged mice [4]. Overall, the human clinical evidence as an administered (topical) therapeutic is limited and inconsistent, and Argireline is not established as equivalent to botulinum toxin [3].
