Summary
FOXO4-DRI is a senolytic peptide that disrupts the FOXO4–p53 interaction to selectively trigger apoptosis in senescent cells, with all available evidence from cell and animal models [1,3]. In aged mice it reduced senescence-associated secretory phenotype output from Leydig cells, improved the testicular microenvironment, and alleviated age-related testosterone insufficiency while improving spermatogenesis and sperm quality [2,3], and in keloid fibroblasts it promoted p53-driven apoptosis of senescent cells, suggesting potential against keloid relapse [1]. Importantly, one model caution exists: in pulmonary vascular disease, eliminating senescent cells (including with FOXO4-DRI) worsened pulmonary hypertension, indicating senolysis is not uniformly beneficial [4]. There are no human interventional trials in the provided literature, so evidence for FOXO4-DRI as an administered therapeutic is preliminary and entirely preclinical.
