Summary
Pasireotide is a multi-receptor somatostatin receptor ligand approved as a prescription therapy, with a monthly long-acting intramuscular formulation approved for both acromegaly and Cushing's disease [1]. In acromegaly it serves as an adjuvant option after surgery, with ongoing efforts to identify biomarkers and an acute test to predict responders [2,3], and in Cushing's disease it provides biochemical control and additionally reduces tumor volume, having been evaluated in high-quality studies [4]. Its efficacy has been demonstrated from preclinical work through phase III clinical trials [3]. The most clinically important safety signal is hyperglycemia, driven by modest suppression of the incretin effect, while cardiac valve disease and psychiatric effects also warrant attention; overall the drug is regarded as well tolerated [1,3,4]. The human evidence is strong and supports pasireotide as an established approved therapeutic.
