Summary
Triptorelin is a gonadotropin-releasing hormone (GnRH) agonist, available as long-acting depot/embonate formulations, and is an FDA-prescription drug used for hormone-responsive conditions [1,2]. In advanced prostate cancer, depot formulations achieve and maintain castrate testosterone levels in the large majority of patients (over 90% maintained castration with the 6-month formulation), are generally well tolerated, and improve quality of life and cost via slow-release dosing [2,4]. For endometriosis, clinical studies demonstrate efficacy in improving pain symptoms and reducing endometriotic nodule volume [1]. In children with central precocious puberty, triptorelin depot suppresses the pituitary-gonadal axis and pubertal development, with quarterly and monthly formulations giving similar results and no reported long-term adverse effects on reproductive function, though some questions on hyperandrogenism/PCOS risk remain to be clarified [3]. The human evidence base across multiple indications is well established.
